Ole Lykke Madsen was born on 14 February 1815, in Holmstrup, Skippinge, Holbæk, Denmark as the son of Christian Jacobsen Lykke and Johanne Larsdatter. He married Ane Jensen on 21 July 1837, in Jyderup, Holbæk, Denmark. They were the parents of at least 2 sons and 4 daughters. He lived in Jyderup, Tuse, Holbæk, Denmark in 1845 and Vig, Ods, Holbæk, Denmark in 1855. He died on 24 October 1856, in Rock Creek, Albany, Wyoming, United States, at the age of 41, and was buried in Atlantic City, Sweetwater, Wyoming, United States.
