When James Thompson I was born on 16 March 1700, in Kilmarnock, Ayrshire, Scotland, his father, Peter Thompson I, was 45 and his mother, Bessie Adam, was 38. He married Agnes Findlay on 31 May 1721. They were the parents of at least 3 sons and 6 daughters. He died in 1748, in Augusta, Hampshire, Virginia, British Colonial America, at the age of 48.
